Moïse was born Martine Marie Étienne Joseph on June 5, 1974, in Port-au-Prince, Haiti. She completed her elementary and high school studies at Roger Anglade School in Port-au-Prince in 1993. She then received a degree in interpretation studies at Quisqueya University in 1997.
She met her future husband, Jovenel Moïse, while both were students at Quisqueya University. The couple married in 1996. Later that same year, Moïse and her husband moved to Port-de-Paix, Nord-Ouest department, with the intention of working in rural development.
During her tenure as first lady, Moïse served as president of the Fondasyon Klere Ayiti, a community development organization focused on civic education and women’s issues. In October 2017, she became president of coordination for Global Fund in Haiti – CCM, which aims to alleviate HIV/AIDS, malaria, and other public health diseases in Haiti. Moïse also advocated for new investments in the Haitian arts and crafts industry in an effort to boost local artisans.
On July 7, 2021, President Jovenel Moïse was assassinated, while First Lady Martine Moïse was shot and seriously wounded during an attack at their residence in Pétion-Ville. Moïse was hospitalized following the attack. Some sources have reported that she died from her injuries later that morning, though that remains unclear.

On 7 July 2021, Moïse was killed when unidentified gunmen attacked his residence in Pèlerin 5, a district of Pétion-Ville. Martine Moïse, the First Lady of Haiti, was hospitalized due to wounds sustained during the attack. A press release issued later that day from the office of acting Prime Minister Claude Joseph blamed the attack on “a group of unidentified individuals, some of whom spoke in Spanish”.
